How do I compute initial spare requirement( say 2 years) for a fleet of 100 vehicles, based on consumption (of spare) per month data, both in range and depth in an optimum way?

1. Determine expected number of failures for the item. [Life data analysis techniques may be used to accomplish this (e.g. assuming a Weibull distribution and instant replacement over a period of time this can be obtained by integrating the failure rate function (e.g. Weibull failure rate function) over the desired period of time. (See Reference 7 at http://www.reliasoft.com/Books/index.html per page 193))
2. In order to correct each failure then a spare needs to be present.
3. Now if you want to optimize, then one needs to decide what to optimize on. The most obvious optimization factor is costs. Thus, one could optimize based on multiple cost variables such as cost of acquiring the spare, storing the spare etc. as well as costs of not having the spare when needed etc.
Hope this helps.

You must have to look the erlang distribution for getting the number of spare part for the whole fleet.
there will be three basic requirement for solving your problem.
1. Basic failure rate.
2. No. of failure per vehicle.
3. Time period as two year in your case.
just follow erlang distribution case and you will get answer.
Hope this will help u .
